Temporal Transcriptional Profiling in response to cMYC and KrasG12D Expression and Inactivation in In Vitro Cultured Primary Mammary Epithelial Cells of Adult Virgin Mice

Mammary glands from 8 week old, female, virgin GEMM MMTV-rtTA/TetO-MYC/TetO-KrasG12D mice were dissociated into single cells and seeded into matrigel matrix for in vitro culture. To determine the gene expression changes which occur upon oncogene induction 5 days after seeding doxycycline was added to the media for 5 days, and then removed. Samples were taken of the following populations: 5 days on, 5 days on/7days off and never induced.
